The larvae were cleared with Fructose until complete bleaching. All samples have been cleared in Fructose solution for several days. Prior to imaging the sample was brought into PBS for several days, embedded in Low melting agarose, the sample chamber filled with PBS and imaged with the 5x detection optics. Each image was obtained by multiview imaging of 5-7 views. The Beads (size 1 µm) for landmark alignment were in the channel with the longer wavelength. The channel was deleted after fusion. Dipl.
